A Straightforward Method to Produce Multi-Nanodrug Delivery Systems for Transdermal/Tympanic Patches

Summary

Transdermal patches have never quite fulfilled their promise as an alternative to injections and oral dosing. This study offers a proof of concept for a dual drug-loaded patch produced with a single piece of equipment: the electrospinning apparatus is used first to spin fibres and then, by tuning the process, to generate nanoparticles that decorate them. Fibre meshes of poly(3-hydroxybutyrate-co-3-hydroxyvalerate) loaded with dexamethasone were combined with rhodamine-loaded PLGA nanoparticles acting as a second model drug. The resulting nanoparticle/fibre systems are intended for local release through the skin and through the tympanic membrane, and were assessed on human dermal fibroblasts.

Electrospinning Starter Kit with Easy Drum rotary system
Both the fibre meshes and the nanoparticles were produced within a single Starter Kit electrospinning apparatus (Linari Engineering S.r.l., Pisa, Italy), using a grounded static collector at 28 cm, together with the Easy Drum rotary unit. Producing fibres and particles on one machine is precisely what the modular design is intended to allow.
